The Northern Adelaide Local Health Network, which includes the Lyell McEwin and Modbury Hospitals, have reintroduced mask wearing for all staff and visitors.
It comes as the state endures another wave of the virus, with reported cases tripling since early October.
"Last Friday it was 50 per cent higher than the previous week," South Australia's Chief Health Officer Nicola Spurrier said today.
The new mask rules apply in all clinical settings including emergency departments and wards, but not in public areas like hospital cafes or shops.
Rising COVID-19 case numbers are already having an impact on healthcare workers.
"We're seeing the furloughing of staff numbers increasing as well - many of those staff will be getting it from the community," Minister for Health and Wellbeing Chris Picton said.
As the festive season approaches authorities are concerned about the elderly, with only half of aged care residents are up to date with their booster shots.
Other hospitals may follow suit and introduce mask mandates, and the situation is being monitored daily.
"Consider wearing a mask in crowded indoor areas and be kind to people who choose to wear a mask," it said on X, formerly Twitter.
It also encouraged people to meet outdoors, wash hands regularly, and stay up to date with COVID-19 vaccinations and called on those feeling unwell to stay at home.
