An unknown shooter opened fire on Ahmed Al Azzam, 25, who was sitting in a car on Mayvic Street in Greenacre, shortly after 2.15am on Sunday.
Less than 50 metres up the road, young couple Kaashif Richards, 22, and Achiraya Jantharat, 19, were sitting in another parked car that was struck by the gunfire.
CCTV from the industrial street shows a dark car speeding away from the scene at the time of the incident.
Detective Superintendent Danny Doherty called it a "brazen, tragic shooting of three people".
"We can't find any links to organised crime conflict or criminal activity," he said.
"The motives are still unknown."
It remains unclear why all three were visiting the industrial street in the middle of the night.
Paramedics treated Al Azzam, Richards and Jantharat at the scene before they were rushed to hospital.
Al Azzam was shot in the head, while Richards was shot in the neck.
Both remain in a critical condition, with Al Azzam fighting for his life.
Jantharat was shot in the back and is in a stable condition.
Superintendent Simon Glasser said multiple shots were fired into both vehicles before at least one shooter fled the scene.
"It's terrible behaviour," Glasser told reporters on Sunday morning.
"The investigation is in its infancy, but we do believe it is a targeted attack.
"It's a really serious offence.
"People shooting at people, it could have resulted in a death at this stage and it still may.
"We treat it at our highest and we will put all of our resources on it."
Police are also probing if a car fire on Amy Street at Regents Park is linked to the shooting, which was reported five minutes after the initial incident.
The car fire spread to a wooden fence and tree and Fire and Rescue NSW crews attended to the blaze.
Crime scenes were established in Greenacre and Regents Park as police began investigations.
Detectives are appealing for anyone who was in the vicinity of either location between 2am and 2.30am and has information or dash cam vision they believe to be relevant to the investigation to contact Auburn police or Crime Stoppers on 1800 333 000.
